Hi Rachit,
You should have mentioned the name of the entrance exam or college about whose application form you are asking for. On a generic scenario, it is seen that always first preference is given if it is available. It never happens that second preference is given when first is available.

Chances of first preference is more. You will get second preference when first is not available.
