Solution : There is error in the "b" part, therefore the correct option is 2.

We need to replace ‘making’ with ‘joining’ or ‘participating in’.

Since a conference cannot be created, anyone can join or take part. Making a conference is therefore useless.

Hence the correct sentence is 

The renowned speaker expressed his helplessness in joining the conference to be held in the university premises.
