Nope! NTA used to allocate the seats based on the preferences you have put in. Now if you have put Kota as you first preference then you have the highest possibility to get a exam seat there if all the seats are not blocked. If all the seats are blocked then they will allocate you seat in your 2nd preferences and the process goes on like this.

For most of the cases, students used to get seats in 1st preference. Its very unusual to get seat allocated based on your 2nd preference.
