The GATE exam is conducted for engineers who have graduated with B.E. or B.tech

So after you have completed your UG you can write the GATE examination.

GATE exam score is required for taking admission to the post graduate courses in engineering descipline such as M.E. and M.Tech.
Also some of the government as well as private organizations prefer GATE score for their recruitment process.
However, the eligibility criteria for GATE is kept as the accomplishment of B.E.or B.Tech degree, so you have to apply for GATE whenever you complete with your graduation.
