Lock and Key hypothesis was given by Emil Fisher . According to this hypothesis, substrates fit into active site of enzyme as key fits into lock.

Induced fit hypothesis was given by Koshland . This hypothesis states that binding of substrate with enzyme causes conformational change of enzyme that enhance its functionality.

These two hypothesis try to explain working of enzymes in biology.
