Definitely you are allowed to give JEE Main exam.But if you want to take admission as lateral entry then you have to clear Jee Advance also with good rank after clearing you Jee Main as there are no lateral entry scheme available for NIT.
I would suggest you to give LEET exam.
LEET is an entrance examination conducted for students seeking admission direct admission to 2nd year of engineering programs through lateral entry offered by various government and private technical Engineering institutes in India.

Yes, definitely you can appear in joint entrance examination after completion of your Polytechnic. But I'll suggest you to appear in Lateral Entry Entrance Test examination to get admission directly in 2nd year of engineering course as this will save your year.
