Hey,
You can give JEE after diploma as well. But then you will not get Lateral entry ( direct entry from 2nd year) in Degree college.
So if you want to go for computer engineering or computer science, then you can give JEE and get in the field you want.
Criteria for JEE for diploma students are as follows:

• Candidate must have completed 10+2 or equivalent examination from a recognised board.
• Candidate must have studied and passed physics and mathematics in 10+2 or diploma examination as compulsory subjects and biology/biotechnology/chemistry as elective subject.
• There is no age limit for eligibility.

You won't be eligible for NITs and CFTIs.

Also, LEET examinations are conducted by various states for diploma holders and it doesn't affect your career negatively. It is a official examination which will give you direct entry in 2nd year of Engineering courses. So, you can definitely go for this examination.
B.Tech after lateral entry and regular B.Tech are same. Infact, job opportunities are also similar for both.
Diploma holders, infact, have an advantage of having more industrial or practical knowledge.

The best colleges for CSE are IITS, BITS Pilani, DTU, college of engineering, Pune, Vellore institute of technology.
