Your brother's 12 marks doesn't decide that if he is eligible for nit or not..it depends on the performance in jee mains exam..if he prepares in this 1 year well..and if his performance is good then...he will surely get nit..

Since there is an eligibility of 75% in board 10+2 is necessary,i would suggest your brother to write improvement exam and get 75% or more and now he will be eligible for NITs.

Yes he's eligibility for jee main if he drop one year as eligibility criteria is Candidates appearing for 10+2 or equivalent exam in 2019 are also eligible to apply irrespective of their age. Candidates must have passed class XII or qualifying exam in 2017 or 2018. Candidates appearing in class XII or equivalent qualifying exam in 2019 can also apply  main.
