Generally when you dont get colleges in first round then you should go for second round to opt for desired colleges.Since you have not mentioned the exam name it is very difficult to say whether there is a third round or not.So you have to lock your desired colleges previously.Rest completely depends upon the counselling process whether your rank  matches with the criteria etc.Hope it helps!!
