No diploma student cannot apply for IIT.

After completing class 10 or class 12, many students pursue a diploma in engineering. Few individuals, however, choose to complete their studies after completing their diploma, and some institutes provide lateral entry with direct admission into the second year of B. Tech for diploma holders. In the case of Indian Institutes of Technology there is no option for special entry into IITs following a diploma, thus candidates must take the JEE Advanced exam, which is the only route to get into IITs for undergraduate engineering courses.

see iits has no admission process for lateral entry student to pursue b.tech at second year. however you need to appear in jee main exam , once you qualify for jee main exam you need to appear in jee advance which is the only entrance test to admission for b.tech course in iits.

students who has completed A Diploma course recognized by AICTE or a State board of technical education of a minimum of 3 year duration are eligible to apply for jee main.
