Yes — if you’ve reported to the Round 1 college, paid the fees, and then get an upgraded seat in Round 2 of MCC NEET UG counselling you will have to join the upgraded college and complete reporting there.
The first college is required to refund your fees, after deducting the caution deposit or any minimal processing charges, as per MCC rules. The refund process usually takes a few weeks and is handled directly the first college to your bank account.

If you’ve reported to the Round 1 allotted college under MCC NEET UG counselling, paid the fees, and opted for upgradation, then yes if you are allotted a new college in Round 2 and take admission there, you are eligible for a refund from the Round 1 college.
