The College was founded in July 1939 by the Government of Bengal and named after Lord Brabourne's wife, who served as Governor of Bengal from February to July 1939.

The College allocated half of its seats for Muslim women and the balance for other communities such as Hindus, Parsees, Sikhs, and Jains. The hostel was reserved solely for Muslim students. Due to a lack of Muslim candidates, Lady Brabourne College began admitting Hindu pupils.
