No. JEE Mains isn't used.

RV college of Engineering takes admission through KCET (45% seats) , COMEDK Entrance exam (30% Seats) and Management Quota (25% seats). If you belong to Karnataka then you can appear for both exams else appear for COMEDK exam to get admission in this college. ( Source: Careers360)

In the above mentioned colleges you will not be able to get CSE Branch with this rank in comedk but you can try in the other branches.

If you are asking about admission to B.E courses offered by RV College of Engineering, admission to 45 percent of the seats (out of the total seats) are filled by the Common Entrance test conducted by Karnataka Examination Authority (KEA), 30% of seats are filled through common entrance test conducted by private mangements association (COMED-K) and 25% by management. All the seats are filled through merit.

If you are seeking admission in M.Tech programmes, you will have to appear for Graduate Aptitude Test for Engineering (GATE). PGCET test is also conducted by the state authorities for admission to PG courses.

GATE/PGCET (KEA) entrance examination quota (only for Karnataka students) is 80% and Management Quota is 20%.
