Yes, you can do PhD directly after B.Tech. You won't need to undertake a Masters degree in between. However, building a profile for pursuing PhD without Masters is quite difficult, and you will have to work on research projects starting now. If possible, try and publish some research papers and get stellar recommendations from your professors.

You haven't mentioned if you have qualified JRF in which subject, anyway in case of University of Hyderabad those who want to pursue PhD in Biochemistry, Plant Sciences, Animal Biology, Biotechnology, Systems and Computational Biology, Mathematics, Statistics, Computer Science, Physics, Chemistry, Public Health have the option to not opt for entrance test, in this case they are 52.5% marks in lieu of written test, however if you choose to appear for the test, then whicherver has highest marks among JRF or written test, that will be taken into consideration. As for other subjects, candidates have to appear for the entrance test, I've provided you all this information on the basis of this year (January)PhD admission process.
