No, you don't have to make registration again. Also, you can't choose any institute once you have locked choice. You will have wait for the second round counselling.

There is no need to register in 2nd round of counseling. You need to register only once and participate in other rounds untill you lock a seat.

You have to accept the seat to be considered for the next round and pay the fees. Onlly after paying seat acceptance fee, you will get willingness to float or freeze. Those who dont pay the fees and accept the allotted seat then they will be out of the counselling
