The eligibility criteria through TSICET includes min. 45% score in Graduation and a good rank in TSICET exam. You have a good rank and if you have also scored min. 45% in your graduation then you are definitely eligible for a seat in a reputed college.

According to neet counselling rules you should have that state domicile certificate to participate in their counselling for 85% quota.
To get a domicile certificate you should have land or house in that state
So if you have punjab's domicile then you can participate in its state quota counselling.
