The eligibility criteria says that that a candidate should be a permanent resident of Assam, so a candidate from other state is not eligible for this examination, this examination is only meant for Assam students.
Further, 2021 has already passed so I will advise you to focus on 2022 examination and you are eligible for Cuet, JEE mains and advanced so start preparing for them.

The eligibility criteria for cee examination is as follows :
Age Limit - 17.5 - 23 years
Educational Qualification - Minimum Qualification of 10+2 pass with specific subject requirements as per the service category
Nationality - Must be an Indian citizen

eligibility to get admissions in the top the BTech Biotechnology colleges, students will have to secure 55% marks in 10+2 with science subjects like Physics Chemistry, and Maths. Some of the entrance exams for BTech Biotechnology are JEE Main, WBJEE, JEE Advanced, KCET etc.
Most private colleges and uiversities take admission in engineering without entrance exams. Though entrance exams are conducted on a national level, state level & university level for BTech admission. But it won't be possible for you to appear in all of them.

Yes, if you have a seat on 2nd round of seat allotment then your first seat will be rejected manually. But, in some case if you have paid your seat booking or admission fee in your first alloted seat then for the 2nd some universities give you the chances to choose between past and present.
