no it is not mandatory to do computer science for diploma in computer science. you can take any diploma without having any previous degree in computer science, but having a bit of basic knowledge in computer is an added advantage and can be done on your interest

It is ok to to MCA after normal B.Sc Graduation with PCM combination. At the time of graduation , it would be  better to do online courses on basic fundamental topics related to computer sc like programming knowledge , DBMS, DSA etc.
