No, it is not compulsory to choose subjects of your graduation only. You can choose any subject of your choice in the Mains exam. There are 25 subjects available from which you can choose. You'll get BSc Major in any subject if you complete 48 credits in the chosen subject as per the norms of IGNOU, for instance if you complete 48 credits in physics you'll be awarded with BSc Major in Physics and the same will reflect in your marksheet and degree
