There are many colleges which offer direct PhD after BTech , you just need to qualify the entrance exams. Or other way is , you can do dual degree from any IIT : Mtech + PhD. The IITs which offer direct PhD are IIT Kanpur, IIT Gandhinagar, etc.

In case of reputed government institutions (viz. NITs and IITs), Ph.D. is a must for getting shortlisted and eligible for the selection process for the post of Assistant Professor.
Some colleges also ask for experience, while others look at your curriculum and projects, others at research publications, research vision
