To get admission in NIT or IIT, you need to complete your class 12 with minimum 75% if you belong to General, OBC or EWS category. Or minimum 65% in class 12 if you belong to SC/ST category.
So first complete your 12th grade and then you must appear for JEE Mains and JEE Advanced exam to get an admission in NIT or IIT.
And you must also be among top 20 percentile of successful candidates of 12th grade exam.
By the above said criterion, you can obtain admission in NIT, IIT.

If you are pursuing your master’s from IGNOU (January 2025 batch), you will become eligible to get into M.Phil. or Ph.D. programme only after completing your master’s degree successfully with the required minimum percentage which is (usually 55%).
However, you should know that as per the latest UGC regulations, M.Phil. programmes have been discontinued in most of the universities across India, and admissions are now being offered directly to the Ph.D. programmes based on NET/JRF scores or university entrance tests.

Masters from IGNOU January 2025, you probably will be done with it late 2026 or early 2027. Still, M.Phil won't be an option as per the new education policy (NEP 2020), it has been abolished.
A great majority of universities including IGNOU have already ceased M.Phil courses. In fact, after your masters completion with minimum 55% marks (or 50% for a reserved category), you are eligible to submit your application for doctoral research directly, not M.Phil. Yes, you can apply for an integrated at IISc but only if your B.Sc. in medical laboratory technician is recognized as a science degree (biological or chemical sciences) and should include subjects like:- biology, chemistry, or biochemistry.
IISc may not accept you if your course is more technical or training based. So it would be better if you do M.Sc. in a science subject and then apply for a regular PhD.

The new 4 year graduation scheme for any degree is just to convert your degree into a Honours, (eg: B.com to B.com Hons). And no, its not compulsory to do a 4 year graduation. You can just go for your Bachelor's degree only. That's totally on you.
