No! you cannot register directly for the round 2 NEET state counselling without participating in the round 1 counselling.
Candidates who do not get any seat allotted in the round 1 counselling automatically gets register for the round 2 counselling. Also, in some cases, the students who gets seat allotted in the round 1 but do not wish to join the allotted seat, needs to register themselves for the round 2 counselling.

No, If you are not registered for round 1 counselling than you are not eligible for 2 round. Assuming that you are asking about NEET counseling, then you are not allowed to do registration in second round counseling. Only candidates registered before 1st round counseling, will be eligible to participate in 2nd round counseling.
