Disadvantage of plasmid pBR322 - Recombinant selection via antiobiotics inactivation is a cumbersome procedure as it requires simultaneous plating on two plates having different antibiotics.
This disadvantage has been overcome via the use of plasmid pUC38 which identifies recombinant in a single step, i.e., plating cells onto agar medium containing ampicillin and X-gal (chromogenic substrate for beta-galactosidase enzyme encoded by lac Z).
