May i again fill up the jee mains forms April with fresh condidate

Well, basically students gets benefit of they not fill the fresh like it may happen with many student that in January they got more marks and in april it goes down so these type of student get benefit because the better result would be considered, so you should think why you want to be fresh candidate, you didn't mention any reason.

If you have filled the jee application form in january than you need not to fill it again, but you need to register for april jee main.

And if you have not attempted jee main in january than you need to fill fresh form.

And if you fill the form twice than there is  chance for cancellation of your application.
