Yes, you are eligible to appear in JEE Main examination after completion of your 12th standard from NIOS Board.

Candidates who have completed 12th standard with Mathematics, Physics and Chemistry subjects from a recognized board are eligible to appear in JEE Main examination. Only candidates who have appeared, qualified and secured top 1,50,000 rank in JEE Main examination are eligible to appear in JEE Advanced examination.

Yes, anybody can give jee mains exams. You are eligible for giving entrance exam. But your passing certificate should be from some recognized school.
