The experimental proof for semiconservatio replication of DNA was first shown in a bacteriumand that bacterium was Escherichia coli.
Actually, semiconservative nature of replication of DNA was proved by Mathew Meselson and Franklin Stahl in 1985 in their experiment.
They performed their experiment in the bacterium Escherichia coli and showed that Dna replication is semiconservative i.e half of the parent DNA is conserved in each new DNA molecule.
