When e coli are cultured in a medium where lactose is not available, the lac repressor is produced which binds tightly to the operator, preventing transcription by RNA polymerase. Thisis due to the negative feedback mechanism of lac operon. Lac operon is negative inducible operon which is on in the presence of lactose. To switch it off in the absence of lactose the repressor is produced.
