There is no such restriction on you even if you fill a withdrawal form and claim to not reappear for the exam next year as such a rule would be outright preposterous and arbitrary. As long as you fulfil the rest of the eligibility criteria for JEE mains 2021,

Usually when you are alotted a seat at some institute then they provide you reporting time . You have to report to that college on that date along with all your needed documents.
