Generally for joining you must have to do B. Tech after polytechnic diploma. Or if you're a science student you can go for B. Tech or B. Sc degree.

You can do Diploma in Computer Science . It will be best for getting job in ISRO.

Ashok,  There are various levels at which you can join ISRO and at various designations.

After 12th: You can get into IIST (Indian Institute of Space Science and Technology) for their Undergraduate programmes. The admission is based on JEE Advanced scores. Which means you need to have studied Physics, Chemistry

Yes ,off course you become an Isro  scientist after completing your MTech but you have to appear the test and if you crack the test with interview rounds then only you will able to take join in ISRO .
