I have got 141 rank in met, and if I go to Manipal counselling and want to just secure a seat, do I need to make DD of 325000 and 10000

Hi Amit,
10,000 may be seat booking fees. You should have to pay the fees first after getting a seat in Manipal. Other fees you should pay in admission time.
Also various colleges has their various payment methods. So first you check the payment method their, hope you also pay the fees by Debit or credit cards rather than DD.
