Hello,
You can appear in JEE mains after completion your 12th from a recognised board as well as after doing diploma but remember that you can get in colleges based in jee mains and you will only be eligible for admission in IITs and for that you will have to crack mains and then appear in JEE advance and if you will qualify with a good rank then you can secure admission in IITs.
