Yeah definitely the diploma holder like Polytechnic can able to write the JEE mains exam but they one thing you should keep in mind that diploma holder will can't be able to take the admission in NIT / CFTI / IIIT only they can be able to take the admission in IIT via JEE Advance. So basically JEE mains is the gateway Inorder to reach at the JEE advance and after getting into the IIT their admission will be considered as the 12th pass out candidate.

surely, you can go for JEE MAIN AND ADVANCED but u'll get admission for 1st year. there is no benefit for your diploma degree.If you are not interested in lateral entry then you can go for it,so think and decide. i suggest you to take LEET exam if your interested. plz check the below link for your reference

Yes, you can appear in joint entrance examination after completion of your Polytechnic but as per my suggestion you should appear in Lateral Entry Entrance Test examination to get admission directly in 2nd year of engineering course.
So appear in LEET entrance exam without wasting your one more year.i hope this information helps you.
