Yes, you can get Lateral entry into diploma courses after 12th. You will get into 2nd year of diploma. The eligibility is that you should have completed 12th or ITI from a recognised institute and PCM is required in 12th. Diploma colleges have their own minimum marks requirements but you have to score around 50% atleast to ensure admissions.
The admission is based on entrance examination conducted by the institutes or by merit marks obtained in 12th.

Now, for your 2nd question, if you want to get admission in IIT, then you will compulsorily have to give JEE examination. You have completed your 12th, so either you can give JEE now or after diploma. But after diploma, you won't get Lateral entry in IIT. after Diploma, you have to give JEE and get admission in 1st year of degree only.
