Dear Manatha,
If you have applied for COMEDK UGET 2018, then there is no need to separately apply for PESSAT, as PES University offers admission on the basis of 3 channels - COMEDK UGET, PESSAT and KCET. So, you can get admission at the university through any of the 3 channels. However, if you want to be more specific about the university itself, then you must take PESSAT or else you have to secure the required cutoff in COMEDK UGET 2018 for PES University.
