I passed class 12 in 2016 should I apply for btech in 2021 in gap between 2016-2020 I passed diploma in engineering so can i illigible for btech in 2021

You can apply in state exam of engineering like upsee from which you can get admission as lateral entry students in second year of BTech. And if you done with you 12th and you have a diploma you can apply for jee main as well. But please do check the eligibility criteria once for diploma holders.

I hope you mean polytechnic by diploma. You have two options after diploma (polytechnic) in engineering:
1. Lateral entry- this is through a state level exam. You will give this exam and if you get good enough marks, you will be eligible for the engineering institutes of the states directly in the second year . But you will not be eligible for NITs and IITs.
2. You can appear in the national level exams like JEE for admission into NITs and IITs among all other institutes.
