There are few criteria that you have to look upon.
If you are born in India, then you are a citizen of this country and can appear for your exams as a regular student, i.e.- Non-NRI.However if you wish to avail the NRI quota then with the proof of permanent residence in another country you can apply for it.
The ease of admission is greater with NRI quota however the course fee is more for students entering through NRI quota.

You cannot declare yourself both domestic student and NRI in the same exam as they have only one category box. However, there are few subcategories which are OBC-PWD, SC/ST-PWD. Such cases are not availing two different quotas but they have one category and the other is the subcategory of that category. But this is not the case with domestic and NRI quota. You cannot describe one as the subcategory of the other. Hope this clears your doubt.
