I have a domicile certificate of Uttar Pradesh made in 2016. i verified it on edistrict UP website.Will it be valid for neet 2021 counseling?

As you have mentioned you have made a domicile certificate for neet UG 2021 and it has been verified on the e-district UP website but as per the guidelines you need to follow that.
Domicile certificate will always be accepted if it a recent one. And yours as you have mentioned that you have made your domicile certificate in 2016 so I would recommend you to make a recent domicile certificate to be on a safer side.
