If a candidate didn't apply for round 1 counselling can he/she be able to register/apply for second round counselling?

You can definitely apply for Neet 2 round counselling.
You just have to go through the mcc official website and just fill in your credentials for new registration.
According to counselling rules, if a student was not able to register for the first round then he or she may directly register himself or herself for 2 round counselling.

Yes, you can surely apply for round-2 even if not applied for round-1 for online Aiq or deemed/central universities ug counselling 2019. For round-2, you need to go for new registration and make payments and choices as per your wish. The starting date for the same is from today till 13th July, 2019 till 05:00 PM and choice Filling will start from 12th July, 2019. Payment facility will be available till 13th July, 2019 till 12:00 NOON.

If you are talking about all India quota counselling of NEET then the answer is YES.
Even if a candidate fails to register for first round of counselling, then he or she can make fresh registration in second round of counselling and carry on with the procedure.
